Keywords: TRIS buffer, TRIS-HCl
Abstract
This protocol describes how to make TRIS buffer (TRIS-HCl) at a 1M concentration and a pH 8.0
Guidelines
This protocol describes how to achieve a TRIS buffer at a pH 8.0. If a lower pH is required, i.e. pH 7.4, then add more hydrochloric acid until desired pH is reached

Safety warnings
Read SDS for TRIS base and Hydrochloric acid prior to use.
Add 121.14g of TRIS base to 800 mL of triple distilled H₂O
Stir solution on magnetic plate using magnetic flea until TRIS base completely dissolved
Using a pH meter, adjust pH of solution to pH 8.0 by adding concentrated hydrochloric acid (HCl)
Add triple distilled H₂O to bring final volume of solution to 1 L
Autoclave
