Abstract
Protocol to transform chemically competent Agrobacterium cells
Materials
YEM plates
Water bath
Liquid nitrogen
Add about ~1 µg of plasmid DNA into the 100 µl of competent cell. Freeze the cells by placing in liquid nitrogen.
Immediately thaw cells by transferring to 37 ℃ for 5 min.
Add 1 ml of YEM to the cells and incubate at 28 ℃ for 4 hrs.
Spread the cells (upto 200 µl/plate) on YEM agar with appropriate antibiotic.
Incubate the cells at 28 ℃ for 1 - 3 days to check for transformants.
