Abstract
This protocol details the αS PFF assembly.
Aggregation
Aggregation
Remove monomer stock from Temperature-80 °C and thaw TemperatureOn ice .
Temperature
Dilute stock to desired concentration (typically Amount5 undetermined ) to a final volume of Amount300 µL in autoclavedAmount2 mL centrifuge tubes.
Set thermocycler for continuous shaking (Centrifigation1000 rpm, 37°C ).

Install Amount2 mL plate.
Run PBS and Protein samples. Take Amount2 µL aliquot at 0 hr time point. Run ThioT Assay.
Take Amount2 µL aliquots twice per day for up to Duration120:00:00 ( 72 hours - 96 hours may be sufficient).

Check ThioT Fluorescence.
Aliquot and store at Temperature-80 °C .

Prepare ThioT stock mixture (Amount8 µg ThioT in Amount10 µL milliQ H2O).
Prepare working ThioT mixture, Amount20 µL stock into Amount1 mL PBS.
Dispense Amount200 µL working solution into each Amount2 µL aliquot.
Dispense Amount200 µL per sample into 96-well, black, corning glass-bottomed plate.
Read Fluorescence Intensity (Ex 450 +/-10; Em 490 +/- 10).
