This protocol is for making 1M calcium chloride solution, which is used in the preparation of worm media, like NGM (Nematode Growth Medium).
Materials
Calcium chloride:ReagentCalcium Chloride DihydrateSigmaCatalog #C7902-500G
Filter: Rapid-Flow Nalgene 0.2µm aPES membrane, ref: 564-0020
Vaccum pump: Fisher Brand, ref: FB70155


Weigh Amount7.4 g ±0.05 of calcium chloride dihydrate (CaCl2·2H2O).

Dissolve it in ~40 mL of milliQ water in a 100mL measuring cylinder.
Add a clean stirring magnet, and leave it on a stirrer until all crystals are dissolved.
Remove the stirring magnet: To prevent touching the solution, use another magnet from the outside of the measuring cylinder to slide the stirring magnet out of the cylinder.
Fill up to Amount50 mL .
Filter-sterilize.
Store at 4°C.
We usually use it without problem for up to at least a year after preparation.
