Abstract
Preparation and Administration of Systemic 4-AP
4-Aminopyridine was obtained from Sigma-Aldrich (Cat #A78403), and a stock solution was prepared in saline. Mice (8 weeks old) were given intraperitoneal injections of soluble 4-AP at a low dose of 2 mg/Kg or equivalent saline.
Animals were monitored every 20 min and presented no sign of convulsive seizure behavior. After 3 hrs, animals were anesthetized with isoflurane (VetOne Cat#502017), followed by cervical dislocation. Brains were rapidly removed and drop-fixed in 4% PFA solution for immunohistochemistry or placed on ice for biochemical assays.
