Abstract
Novel object recognition rodent behavioral test protocol
Set Up
Set Up
Clean open field maze with 70% ethanol and obtain 3 sets of different novel objects.
Set up open field maze (Noldus Maze Engineeres) with 4x single arenas with dimensions 40cm x 40cm x 40cm.
Habituate mice to room for approximaetly 1 hour prior to testing.
Obtain two distinct objects of similar complexity (blocks of the same size, different shape).
Level of manipulation of objects by rodents should be minimal and consistent such that objectcs cannot be moved.
You will need two of the same novel objects to train the mice. Example; two blocks pyramind shaped blocks, one rectangular block
Procedure - Training Session
Procedure - Training Session
Mice should be habitutated to the open box without either novel object for 5 minutes.
Position both that identical objects in the open field, allow mice to explore for an additional 5 minutes.
Record the time spent exploring each object.
Exploration should be defined as sniffing, biting, or facing the object.
Clean open field and objects with 70% ethanol between each animal training session.
Procedure - Test Session
Procedure - Test Session
24 Hours after training session, repeat step 5 to habitatute mice.

After this, allow 5 minutes of exploration, time this time using one of the familiar objects and the novel object.
Record the time spent exploring the novel object and familiar object.
Clean open field and objects with 70% ethanol between each animal test session.
