Be aware PSI Closed Fluorcams are configured to customer specifications and can contain different actinic light sources. The provided Fluorcam method protocol is written for the IGB GEGC Fluorcam FC800-331 and assumes that ACT1 is configured with a red light source and ACT2 is configured with a white light source. The Fluorcam determines light intensity as a percent of maximum. The PAR values given in this protocol were determined empirically using a LI-250 PAR meter and assume a 96 well plate is being analyzed on the third shelf from the top.
This protocol has successfully been used on soybean, tobacco, and cowpea. For plant species with highly responsive NPQ such as C4s, the timing of the saturating measurement pulses may need to be altered to get smooth relaxation curves for proper curve fitting.
If modifications are made to the Fluorcam method protocol provided, be aware the maximum run time on a Fluorcam FC800-331 with FluorCam 7 software is ~ 75 minutes.
