Abstract
β-Galactosidase staining
For cell staining
For cell staining
Culture cells on Matrigel-coated coverslips.
Fix cells with the fixation solution provided in the Senescence β-Galactosidase Staining Kit (Cat. number 9860; Cell Signaling Technology).
For tissue and organoid sections
For tissue and organoid sections
Prepare 20 µm OCT-embedded sections by cryosectioning.
Mount sections on Epredia SuperFrost Plus slides (Fisher Scientific).
Refix sections with the fixation solution provided in the staining kit.
After fixation for both cells and sections
After fixation for both cells and sections
Perform β-Gal staining on both cells and sections according to the manufacturer’s instructions.
Acquire images:
  • Use a Leica DM750 brightfield microscope with a 40×/0.4 numerical aperture objective for cells.
  • Use an EVOS M7000 Imaging System with a 20×/0.4 numerical aperture for tissue and organoid sections.
