Add Amount2 mL (Amount3 mL also acceptable) of LB medium and the appropriate antibiotic that selects for the target strain into a 15 mL tube.

Gather cells.
Swab a single colony on the agar plate with a sterile 1µL inoculation loop.

OR Dip a sterile 10 µL innoculation loop in a preexisting culture tube.

Innoculate the 15 mL tube with the loop.
Place the 15 mL tube in the thermomixer for 16 hours at 37°C and 300 rpm.
